Demonstration of rhodanese activity in polyacrylamide gels.

چکیده
Rhodanese activity from crude extracts of Thiobacillus sp. strain IV-85 was demonstrated in polyacrylamide gels after incubation in the reaction mixture by staining with dichloroindophenol in the presence of methylphenazonium methosulfate. The sensitivity of the staining system was found to be 8 x 10 moles of sulfite.

Structural studies of bovine liver rhodanese. I. Isolation and characterization of two active forms of the enzyme.
Crystalline bovine liver rhodanese, prepared by ammonium sulfate and pH precipitation, has been shown to be comprised of two fully active components present in approximately equal amounts which are separable by polyacrylamide gel electrophoresis and by ion exchange chromatography. The two rhodanese forms, designated A and B on the basis of their order of elution from columns of DEAE-Sephadex, a...
متن کاملA Novel Method to Detect β-Cyclodextrin Glucosyl Transferase (β-CGTase) Activity on Polyacrylamide Gels
b-cyclodextrin glucosyl transferase (b-CGTase) hydrolyses starch to produce b-cyclodextrin by transglycosylation (cyclization) activity. The conventional method for detection of b-CGTase activity is based on detecting starch hydrolysis by iodine staining. This method reveals all amylolytic enzymes, but can not discriminate them. In the present study, we introduce a new method for specific detec...
متن کاملProteolytic interconversion of electrophoretic variants of the enzyme rhodanese.
It has been confirmed that the enzyme rhodanese, although a homogeneous single polypeptide chain protein by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, is separable by electrophoresis under nondenaturing conditions into four species which differ in net surface charge (I-IV in the order of increasing positive charge). Limited proteolysis can interconvert these species. Chymotrypsi...
متن کاملUsing a Temperature Gradient against the Time in Polyacrylamide Gel Electrophoresis May Eliminate the Need for Stacking Gels
Background and Objectives: Making stacking gels for polyacrylamide gels in the laboratory by conventional methods is laborious and time consuming. Considering the role of temperature in polyacrylamide gels with respect to electrical resistance and viscosity, we assumed that decreasing the temperature would cause an increase in electrical resistance and viscosity. Ultimately, a downward tempera...
متن کاملCharacterization of PspE, a secreted sulfurtransferase of Escherichia coli
PspE, encoded by the last gene of the phage shock protein operon, is one of the nine proteins of Escherichia coli that contain a rhodanese homology domain. PspE is synthesized as a precursor with a 19-amino acid signal sequence and secreted to the periplasm. Mature PspE is the smallest rhodanese of E. coli (85 amino acids) and catalyzes the transfer of sulfur from thiosulfate to cyanide forming...
